ABSTRACT
Most of the medicines are unable to permeate the skin at the required rates, it has significantly restricted the success of transdermal medication delivery. The main obstacle in topical drug delivery is the skin, providing the opportunities to create the techniques that evade this barrier for the efficient topical delivery of high molecular weight compounds and nucleic acid-based therapeutics. Drug delivery with micro and nanotechnology has been improved to reach the optimum needs of the drug in comparatively smaller doses and with the convenience of administration to the patient. The use of micron-scale needles to promote skin permeability has recently been proposed and demonstrated to greatly boost transdermal delivery, particularly of macromolecules. Using the tools of the microelectronics industry, micro-needles have been prepared having various sizes, shapes, and materials. A microneedle-based drug delivery system is a little invasive administration, user friendly, continuous, and controlled release system. Microneedles are micro-sized needles that are dissolvable and are painless methods of administering the drug into the skin. The microneedles patch that is 100–1000 μm in size pokes through the skin and allows micron scale drugs to cross into the bloodstream. The microneedles transport drugs to the skin deep into the systemic circulation. Micro-needles in a variety of sizes, forms, and materials have been produced using microelectronics industry equipment. The microneedle system can be used for delivery of oligonucleotides, vaccines, insulin, Painless administration, convenience of use, discrete, continuous, and controlled release are all advantages of a microneedle drug delivery system.This review will help to understand various fabrication materials required and different methods used for the development of microneedles. This article will give the distinctive advantages and disadvantages also of microneedles along with a broad description of potential for pharmaceutical and biomedical applications. The mechanism and role of microneedles in serving as a platform to treat various ailments. The review also inculcates the pharmacokinetics, in vitro/in vivo toxicity, biosafety, and marketed product of microneedles. We have also discussed the clinical trials and patents on the pharmaceutical applications of microneedles in brief. In short, this article will give insight into microneedle technology and provide a recent advancement, which pave the pathway for future pharmaceutical and biomedical applications.
